Growth of cultured human epidermal cells into multiple epithelia suitable for grafting.

Abstract: Owing to several recent developments, the cultivability of epidermal keratinocytes, particularly those of the human, has been greatly improved. Under the conditions used, single cultured cells generate stratified colonies that ultimately fuse and form an epithelium that is a reasonable approximation of the epidermis. It will be shown here that large amounts of cultured epithelium can be generated from a small piece of epidermis in a short time. We wish to bring to the attention of surgeons and cell biologists … Show more

“…Primary cultures of human craniopharyngioma cells were isolated and prepared from tumor samples in a similar manner as for keratinocytes according to the methods described (30). Briefly, after surgical removal, a portion of the tumors was immediately put in DMEM with 10% FCS and stored at 4jC.…”

“…Cell Culture-Punch biopsies obtained from the probands were used to establish primary cultures on lethally irradiated murine 3T3 fibroblasts as described earlier (37)(38)(39). Secondary cultures were grown in high calcium keratinocyte medium, 10% fetal calf serum until confluency.…”

“…1 Skin substitutes have the ability to deliver cytokines, chemokines, growth factors and competent cells at wound site, thus improving the wound closure. 2,3 Definitive treatment of extensive burns requires that autologous skin be used since other allogenic grafts are rejected after several days/weeks.…”
