Gut mucosal virome alterations in ulcerative colitis

Abstract: ObjectiveThe pathogenesis of UC relates to gut microbiota dysbiosis. We postulate that alterations in the viral community populating the intestinal mucosa play an important role in UC pathogenesis. This study aims to characterise the mucosal virome and their functions in health and UC.DesignDeep metagenomics sequencing of virus-like particle preparations and bacterial 16S rRNA sequencing were performed on the rectal mucosa of 167 subjects from three different geographical regions in China (UC=91; healthy contr… Show more

“…The total collection of vaginal viruses (the vaginal “virome”), has only been sparsely investigated, but there is increasing evidence that bacteriophages (or “phages”) are a factor in certain diseases related to gut microbiome dysbiosis[9]. Further, phages adhere to and are significantly enriched in mucosal surfaces, possibly providing what has been termed “non-host derived immunity” against infection[10,11].…”

“…Immune-related digestive system inflammation, including colitis, hepatitis, pancreatitis, can be caused by a variety of pathogenic factors, such as genetic abnormality, autoimmune factors, immune-related drugs, viral infections, and so on (1)(2)(3)(4). PD-L1(B7-H1) is thought to be involved in the regulation of cellular and humoral immune responses through the PD-1 receptor on activated T and B cells (5,6).…”

“…For example, there have been more attempts to characterize “healthy gut phageome” since a study in 2016 conducted deep-sequencing of DNA from virus-like particles (VLP) and revealed the presence of completely assembled phage genomes in 64 healthy individuals around the world (8). Attempts have also been made to explore the associations between human gut virome alterations and diseases (7, 9, 10). However, only a small number of metagenomic studies focused on oral phage communities (5), including a study in 2014 that reported an alteration of virome composition in subjects with periodontal disease (11).…”
