Halotolerant Aerobic Heterotrophic Bacteria from the Great Salt Plains of Oklahoma

Abstract: The Salt Plains National Wildlife Refuge (SPNWR) near Cherokee, Oklahoma, contains a barren salt flat where Permian brine rises to the surface and evaporates under dry conditions to leave a crust of white salt. Rainfall events dissolve the salt crust and create ephemeral streams and ponds. The rapidly changing salinity and high surface temperatures, salinity, and UV exposure make this an extreme environment. “…DSM 3050 was isolated from a sample from the North Arm of Great Salt Lake in Utah and grew optimally in the presence of about 10% NaCl (15,37). Both GSP65 and GSP66 were isolated from samples from the great salt plains of Oklahoma, and these strains were recovered together with a collection of 105 isolates belonging to a wide variety of taxa, indicating that their abundance in the cultivable fraction of their community is relatively low (11). It is interesting to note that a previous examination of the tolerance of the 63 GSP isolates of Caton et al (2004) to heavy metals revealed that GSP 10 and 21 were most tolerant of 1000 ppm lead and cadmium, while GSP 26, 30, and 58 also performed well, with GSP 12 tolerant to lead but not cadmium (Bhattarai et al, 2004).
