Heat shock protein 90’s mechanochemical cycle is dominated by thermal fluctuations

Abstract: The molecular chaperone and heat shock protein 90 (Hsp90) exists mainly as a homodimer in the cytoplasm. Each monomer has an ATPase in its N-terminal domain and undergoes large conformational changes during Hsp90's mechanochemical cycle. The threecolor single-molecule assay and data analysis presented in the following allows one to observe at the same time nucleotide binding and the conformational changes in Hsp90. Surprisingly, and completely unlike the prior investigated systems, nucleotides can bind to the … Show more

“…Therefore, our results support previous findings showing that asymmetry plays an important role in homodimeric Hsp90 systems: asymmetry is required for client binding 39 , asymmetric SUMOylation facilitates cochaperone binding 40 , nucleotide affinity is asymmetric 22 and hydrolysis is only required in one subunit 41,42 .…”

“…The cycle for the N-terminal open-close movement has recently been solved 22 . Putting the numbers from this publication into equation (1) we obtain a 'directionality' of around 1 k B T. This is the thermal energy and therefore consistent with the finding, that idle Hsp90 is mainly dominated by thermal fluctuations.…”
