We examined the blood biochemical profile and the physiological response to acute handling stress of a historically important economic but currently a rare endangered fish, Largemouth bronze gudgeon, Coreius guichenoti , in the upper Yangtze River. The results showed that the glucose (GLU) and triglyceride (TG) levels in the cage-cultured C. guichenoti were significantly higher than those of the wild type, and the basal cortisol (COR) level was higher than that of most other fish species. In the stress experiment, the fish were subjected to a low-and high-intensity handling stress, respectively, and sampling was performed before the stress and at 0, 1, 4, 12, 24, 72, 120, and 168 h after the stress. The levels of COR, GLU, lactate (LA), and alanine aminotransferase (ALT) of stressed fish exhibited an increase first and then a recovery to the pre-stress levels with a typical acute stress response in the first 24 h. Fish in both stressed groups were spontaneously infected with Ichthyophthirius multifiliis from the 24th to the 168th h, and exhibited a chronic stress response. These data would facilitate the studies of blood physiology of C. guichenoti and related fish in health evaluation, species propagation and conservation, as well as the stress response in aquaculture.

The aim of this study was to obtain a basic knowledge of the hematology of Schizothorax (Racoma) davidi (Sauvage). The results showed that red blood cell and white blood cell counts as well as mean cellular hemoglobin concentration were significantly higher in females than in males, while mean cell volumes were significantly lower in females than in males. No significant differences were seen in other hematological parameters in relation with gender. Under light microscope, erythrocytes (mature and immature), leukocytes (neutrophils, lymphocytes, and monocytes), and thrombocytes were distinguished. Mature erythrocytes were ellipsoidal cells with oval, central nucleus. Neutrophils contained banded or segmented nuclei. Small lymphocytes had a very thin rim of basophilic cytoplasm. Monocytes often possessed vacuoles in their cytoplasm. Thrombocytes existed in several forms with relatively large nucleus surrounded by a pink-stained or hyaline cytoplasm. The percentage of the different leukocytes revealed predominance of neutrophils and small lymphocytes, followed by monocytes and large lymphocytes. The neutrophil percentages of each stage showed that metamyelocyte accounted for the most, followed by banded neutrophils. This investigation could be used for evaluation of the health status of cultured male and female S. davidi and will provide early detection of clinical pathology.

Structural and functional characterization of peptide YY on feeding in Schizothorax davidi

Several studies have demonstrated that the neuropeptide peptide YY (PYY) plays an important role in feeding in mammals and fish. However, thus far, the feeding regulation function of PYY in Schizothorax davidi has not been well understood. Here, we identified the full-length cDNA sequence of PYY in S. davidi for the first time. S. davidi PYY contains 803 bp nucleotides including a 328 bp 3' untranslated region (UTR), a 181 bp 5' UTR, and a 294 bp open reading frame encoding a peptide of 97 amino acids. S. davidi PYY expression was observed in almost all tissues, with the highest expression detected in the hypothalamus. PYY mRNA expression in the hypothalamus was significantly elevated after a meal (P < 0.01), and significantly decreased after fasting (P < 0.01). PYY expression levels were increased sharply following refeeding after 9 days (P < 0.01), suggesting that it might function as a satiety factor in S. davidi.
