Hepatic differentiation of human bone marrow‐derived UE7T‐13 cells: Effects of cytokines and CCN family gene expression

Abstract: The present study suggests that aFGF/bFGF/HGF/type IV collagen coating is the efficient condition for hepatic differentiation of UE7T-13 cells, and that WISP1 and WISP2 play an important role in hepatic transdifferentiation of these cells.

“…During the second stage, from 9th to 14th day , albumin (ALB), ornithine transcarbamylase (TAT) as well as mRNA levels for VII clotting factor, asialoglycoprotein receptor 1 (ASGPR1), and CYP1A2 member of cytochrome P450 superfamily production are increased while AFP levels are reduced [6568]. The last stage of maturation process, from 15th to 21st day , presents hepatic-like cells with functional properties unique to hepatocytes such as increased detoxifying ability, lipid and glycogen storage capacity, urea production, and bile acid secretion [6568]. …”

“…SH9 human PDL cells were maintained in α-modified minimum essential medium (α-MEM; Gibco, Life Technologies, Gaithersburg, MD, USA) supplemented with 10% fetal bovine serum (FBS; Gibco), 50 µg/ml streptomycin, and 50 U/ml penicillin. UE7T-13 cells, which are human bone marrow-derived MSCs infected with retroviruses expressing papillomavirus E7 and hTERT to prolong their lifespan (13,14), were purchased from the Health Science Research Resources Bank (JCRB1154, Japan Health Sciences Foundation, Tokyo, Japan) and cultured in α-MEM. All cultures were maintained at 37˚C in a humidified atmosphere of 5% CO 2 .…”

“…cells was prolonged by infecting them with a retrovirus containing human papillomavirus E7 and telomerase reverse transcriptase (hTERT) cDNAs (Mori et al, 2005;Shimomura et al, 2007;Ishii et al, 2008;Takeuchi et al, 2007). We first tested the ability of hESF9 medium, which we had developed for use with hES cells, to support the growth of UE7T-13 cells.…”
