Heterogeneous distribution of k13 mutations in Plasmodium falciparum in Laos

Abstract: BackgroundThe emergence and transnational spread of artemisinin resistance in Plasmodium falciparum in the Greater Mekong Sub-region (GMS) is a serious threat to malaria elimination in the region and could present a threat to malaria control in Africa. Recently, the Lao Government adopted the goal of malaria elimination by 2030, for which monitoring of artemisinin-resistant malaria within the country is indispensable. This study’s objectives were to assess the distribution of k13 mutations in Laos.MethodsPlasm… Show more

“…Although significant progress in the fight against malaria has been made, the data reported by the World Health Organization for the period 2015-2017 remain worrying with no reduction in the number of cases worldwide [1]. The resistance developed by the vector to insecticides [2] and the therapeutic failure of antimalarial drugs [3,4] explain the reasons for the current prevalence of the disease. In addition, access to care for populations living in malaria endemic areas remains a critical problem, especially in sub-Saharan Africa [5].…”

“…DNA samples were extracted from three dried blood spots with a diameter of 3 mm using a Maxwell® RSC DNA FFPE Kit (Promega, Madison, WI, USA) and the automated Maxwell® RSC Instrument (Promega). Minor modifications to the manufacturer's guidelines can be found in a previous report [27]. DNA was eluted in 50 µL of elution buffer and stored at 4 °C.…”

“…The regional strategy for malaria elimination aims at eliminating malaria foci in all GMS countries, maintaining a malaria-free status, and preventing reintroduction. The strategy also includes urgent action to eliminate Plasmodium falciparum malaria by 2025 to contain the spread of multi-drug resistance in the GMS and in the southern part of Laos, specifically [ 1 , 3 , 7 ].…”
