HI‐6 and 2‐pam in sheep: Pharmacokinetics and effects on muscle tissue following intramuscular injection

Abstract: The pharmacokinetics of 2-PAM, a component of the current nerve agent antidote therapy for U.S. military forces was compared to the pharmacokinetics of another acetylcholinesterase reactivator HI-6. Additionally, the effects of these compounds on muscle tissue following intramuscular injection was examined. Plasma concentrations of the oximes were determined by HPLC. Plasma concentration-time profiles for both oximes fit a one-compartment open model with first-order absorption and elimination. The results demo… Show more

“…Blood samples (0.4mL) from the carotid artery were drawn 5, 4, and 3min before injection and 0, 1, 2, 5, 10,15,20,25,30,40,50, 60, 90, 120, 150, 180, 210,240,270,300,330, and 360min postinjection. Part of each sample (0-3 mL) collected in heparinized syringes was used for measurements of Pao2, Pacoz, and pH by an ABL 2 MT analyser (Radiometer A7S).…”

“…It has been shown that therapeutic concentrations of 2-PAM are quite high in rat plasma and serum (1.4-5.5 mg/ml; [27] up to 100 mg/ml; [26] 0.7-3.3 mg/ml [28] ), in sheep plasma (up to 7 mg/ ml, [33] and in human plasma and serum (0.5-3 mg/ml; [23,24] 3-6 mg/ml; [25] 3-15 mg/ml [12,13] ). Concentrations in human urine were more than 100-times higher (250-2000 mg/ml [20] ).…”

“…170–206 min). Parameters are species‐dependent as shown for example, for man, rat, and sheep . Elimination of 2‐PAM is best described by a two‐compartment open model not showing significant binding to plasma proteins …”

“…As summarized in Table , diverse IPC modifications were introduced in the following years varying the stationary phase material and mobile phase composition. IPC was applied to qualitative and quantitative analysis of neat solutions, pharmaceutical formulations and biological specimen from different species including plasma and serum, urine, and brain tissue (Table ).…”
