High-Level Production of Bacteriotoxic Phospholipase A1 in Bacterial Host Pseudomonas fluorescens via ABC Transporter-Mediated Secretion and Inducible Expression

Abstract: Bacterial phospholipase A1 (PLA1) is used in various industrial fields because it can catalyze the hydrolysis, esterification, and transesterification of phospholipids to their functional derivatives. It also has a role in the degumming process of crude plant oils. However, bacterial expression of the foreign PLA1-encoding gene was generally hampered because intracellularly expressed PLA1 is inherently toxic and damages the phospholipid membrane. In this study, we report that secretion-based production of reco… Show more

“…To determine the effect of temperature on refolded PLB1 activity, PLB1 and yolk lecithin mixtures were incubated for 15 min (pH 7.0) at different temperatures, ranging from 20 to 80 °C. PLB1 activity was measured by pH-stat technique as previously described (Park et al 2020). The amount of free fatty acids released from a lecithin solution can be quantified by adding 0.01 M NaOH to maintain a constant pH of 7.0 using an automatic pH-stat (Metrohm, Switzerland).…”

“…In the last decades, several PLA 1 genes from microorganisms had been cloned and expressed and the most frequently studied PLA 1 genes were from Serratia sp. strains [9][10][11][12]. However, PLA 1 secreted by Serratia sp.…”

“…A wide variety of proteins are secreted through this oligomeric secretion channel to play their biological roles. Due to the simple structure of the system, T1SSs have been widely applied in biomedical engineering applications ( Schwarz et al, 2012 ; Ryu et al, 2015 ; Park et al, 2020 ).…”
