High performance liquid chromatographic determination of subpicomole amounts of amino acids by precolumn fluorescence derivatization with o-phthaldialdehyde

Abstract: Using reversed-phase high performance liquid chromatography, 25 amino acids as o-phthaldialdehyde derivatives were determined with a 30-min sample turnover. The derivatization procedure is rapid, performed in aqueous medium, and has few transfer steps. The precision is ±0.5% at the 80-pmol level. Recoveries of standards added to urine samples were >98%. Intercallbration with an amino acid analyzer showed an agreement of >95 %. The detection limit Is about 50 fmol. The overall retention is controlled by the org… Show more

“…Endogenous glutamate analysis GLU was measured by precolumn derivatization with o-phthaldialdehyde and separation by reversed phase HPLC coupled to fluorimetric detection (Lindroth and Mopper 1979), according to the method described by Calò et al (1997) and modified by Morari et al (1998). Fifteen microlitres of o-phthaldialdehyde/mercaptoethanol reagent (0.4 M borate, 0.04 M o-phthaldialdehyde, 0.4 M 2-mercaptoethanol, pH 10.4) were added to 15 lL aliquots of sample.…”

“…Vol. 5, No.3, 1985 Amino acids were measured with liquid chromatog raphy after precolumn derivatization with o-phthaldialde hyde (OPA) (Lindroth and Mopper, 1979), The OPA derivatives were separated on a 5 fLm Nucleosil C-18-column (x 200 4-6 mm) using gradient elution with increasing methanol (30-100%) in Na-phosphate buffer (50 mM, pH 5,25). The system consisted of a Varian LC 5000 chromatograph, a Schoeffel FS 970 fluorimeter, and a Waters WISP 710 B autoinjector equipped with an ad ditional pneumatic valve to perform automatic derivati zation and sample injection into the main stream without interruption of the normal flow.…”

“…The ammonium concentrations of this supernatant as well as of the culture medium obtained in the first centrifugation step were determined by reversed-phase highperformance liquid chromatography (HPLC) using a protocol based on the method described by Lindroth and Mopper [21]. Ammonium was labeled by pre-column derivatization using ortho-phthaldialdehyde (OPA).…”
