High-Pressure Liquid Chromatographic Assay of Netilmicin in Plasma

Abstract: A high-pressure liquid chromatographic method for the quantitative determination of netilmicin in plasma was developed. The procedures involve acetonitrile protein precipitation, methylene chloride extraction, and dansylation to form the fluorescent dansyl derivative of netilmicin, which is extracted into ethyl acetate and chromatographed on a reverse-phase column with aqueous acetonitrile as the mobile phase. A good linear relationship between peak height measurements and netilmicin concentrations was found. … Show more

“…Urinary recovery over the 10-day collection period was greater than 94% of the administered tobramycin dose for two of the subjects, whereas only 83% of the dose was recovered from the third subject, probably due to his failure to collect all urine. DISCUSSION A number of high-performance liquid chromatographic assays have been developed for the analysis of aminoglycosides in serum (1,2,6,7,10). The present method is comparable in specificity and improved in accuracy.…”

“…RESULTS AND DISCUSSION Recently, HPLC methods have been used widely to determine the plasma and serum concentrations of antibiotics such as netilmicin (6) Figure 1 shows the typical liquid chromatogram of Sch 29482 and the internal standard (m-propionamidophenol) extracted from control serum and from the serum spiked with Sch 29482. The retention times for Sch 29482 and the internal standard were 8 and 10 min, respectively.…”

In this report, we describe the application of HPLC for the separation and detection of amikacin.…”
