High-resolution laser desorption mass spectrometry of peptides and small proteins.

Abstract: Matrix-assisted laser desorption/ionization (MALDI) has been used with an external ion source Fouriertransform mass spectrometer to obtain the highest mass resolution ever, to our knowledge, demonstrated for laserproduced ions (r/Am = 1,100,000 for [Arg8Jvasopressin, 228,000 for melittin, and 90,000 for bovine insulin). The peaks in the isotope cluster for bovine insulin are fully resolved, and the mass measurement accuracy is an order of magnitude better than can be achieved with time-of-flight mass spectrome… Show more

“…Dried samples were dissolved in 0.02 N NaOH and purified by HPLC. The purified product was analyzed by T 2 S RIA and high-resolution mass spectrometry (MS) (9,10). RIA analysis revealed a T 2 S-reactive peak, the elution time of which coincided with W peak on HPLC.…”

“…Examples of these efforts include the use of a comatrix [2] or a "waiting room," [3] which in the limit becomes high-pressure MALDI [4]. An alternative is the use of an external source [5], which permits the use of quadrupole and higher order traps or ion guides to focus and thermalize the MALDI-produced ions before introducing them into the FTMS trap. The second approach is to introduce efficiently the MALDI ions, whether produced externally or in-field, into the trap.…”

“…The image current produced by the motion of the coherent ions is then recorded and Fourier transformed to produce a mass spectrum. In comparison to other mass analyzers, FTICR-MS has the capability of analyzing high mass bio-molecules with high sensitivity, resolution and mass accuracy (McIver, Li, & Hunter, 1994;McLafferty, 1994;Kelleher et al, 1995). Therefore, it has attracted a wide range of applications in biological sciences from genomics to metabolomics in spite of the high cost of FTICR-MS instruments.…”
