Highly cytotoxic DNA-interacting copper(ii) coordination compounds

Abstract: Four new Schiff-base ligands have been designed and prepared by condensation reaction between hydrazine derivatives (i.e. 2-hydrazinopyridine or 2-hydrazinoquinoline) and mono- or dialdehyde (3-tert-butyl-2-hydroxybenzaldehyde and 5-tert-butyl-2-hydroxyisophthalaldehyde, respectively). Six copper(II) coordination compounds of various nuclearities have been obtained from these ligands, which are formulated as [Cu(L1)Cl](CH3OH) (1), [Cu(L2)NO3] (2), [Cu2(L3)(ClO4)2(CH3O)(CH3OH)](CH3OH) (3), [Cu2(L4)(ClO4)(OH)(CH… Show more

“…On increasing the concentration of CT-DNA, hypochromism of 6.34 % and redshift of 9 nm for 1, and hypochromism of 14.42 % and redshift of 12 nm for 2 are observed. Hypochromism with redshift is pointed out interaction of DNA via ligand intercalation [47]. This kind of findings were observed in another cupperscorpian like ligands [48] However, complex 3 shows hyperchromic effect (18.25 %).…”

“…Cu(II) are well known to be a redox-active metal whose complexes can generate reactive oxygen species (ROS) which trigger DNA degradation and apoptosis of cancer [7][8][9]. Schiff bases copper(II) complexes have been extensively studied as encouraging alternatives to cisplatin and anticancer drugs [10][11][12].…”

“…Table-1. From this spectral data, it is found that g|| (2.24) > g⊥ (2.03) > ge (2.0024), which revealed that the one unpaired electron is present in (dx2-y2) orbital and it is likely to be the octahedral geometry as conferred in electronic spectroscopy [46]. The g|| value of dimeric Cu(II) complex can be used as a measure of covalent behaviour of copper-ligand interaction.…”
