Highly sensitive and rapid detection of SARS‐CoV‐2 via a portable CRISPR‐Cas13a‐based lateral flow assay

Abstract: To rapidly identify individuals infected with severe acute respiratory syndrome coronavirus‐2 (SARS‐CoV‐2) and control the spread of coronavirus disease (COVID‐19), there is an urgent need for highly sensitive on‐site virus detection methods. A clustered regularly interspaced short palindromic repeat (CRISPR)/CRISPR‐associated protein (Cas)‐based molecular diagnostic method was developed for this purpose. Here, a CRISPR system‐mediated lateral flow assay (LFA) for SARS‐CoV‐2 was established based on multienzym… Show more

“…Liu et al developed a CRISPR-Cas13a-based immunochromatographic assay and designed three-line LFIA test strips for SARS-CoV-2 detection. 114 This method obtained comparable detection levels to RT-PCR assays with a LOD of 0.25 copies per μL and sensitivity of 100%. It should be noted that the CRISPR–immunochromatographic method allowed the test results to be reported within 1 h, thereby saving more than 30 min compared with RT-PCR.…”

“…Fluorescence-based immunoassay technology has attracted considerable attention owing to the development of numerous nanomaterials with good luminescence properties, 115 including quantum dots 116 and carbon dots. 117 However, most fluorescence immunoassays are based only on changes in the fluorescence intensity, which are susceptible to external interference. Liao et al 118 used PBNPs to bind goat antimouse antibodies to obtain a PBNP-antibody binding ratio fluorescence reading, which was then calibrated for different concentrations of histamine.…”
