Highly sensitive colorimetric enzyme-linked oligonucleotide assay based on cyclodextrin-modified polymeric surfaces

Abstract: In this paper, we describe the development of an enzyme-linked oligonucleotide assay for the detection of a human leukocyte antigen allele associated with celiac disease based on cyclodextrin-modified polymeric surfaces. The surface of maleimide-pre-coated plates was modified with a layer of thiolated cyclodextrin polymer and used for the supramolecular capture of adamantane or ferrocene-modified carboxymethylcellulose polymers bearing DNA probes. The assay was optimised in terms of incubation time, temperatur… Show more

“…Horseradish peroxidase (HRP) is one of the most common enzymes used in these assays, in various bioconjugate formats with DNA probes, nanoparticles or other molecules (Merkoçi, 2010;Wang, 2003). Direct conjugation of the ssDNA detection probe with HRP (ssDNA-HRP conjugate) has been used in both colorimetric and electrochemical assays as an alternative to biotinylated detection probe in combination with streptavidin-HRP (Civit et al, 2012;Dequaire and Heller, 2002;Ortiz et al, 2012;Wang et al, 2008). The signal generated though is limited since in general a maximum of one HRP molecule can be linked to each target molecule.…”

“…[24][25][26][27][28][29][30][31] Amongst different host-guest pairs, b-CD and adamantane (Ada) are a strong pair and have been widely used as linkers for integration of biomolecules to construct functionalized bio-substrates. [32][33][34][35][36][37] They can also be dissociated for the regeneration of biomaterials and sensor chips. 37,38 Herein, poly(N-isopropylacrylamide-co-1-adamantan-1-ylmethyl acrylate) [poly(NIPAAm-co-Ada)] was used as a platform for attaching molecular recognition functionalities onto a silicon surface via host-guest interactions between adamantane and ligands bearing b-CD (CD(X)) (Scheme 1).…”

“…Supramolecular host-guest chemistry has recently emerged as a versatile entry for the reversible immobilization of biomolecules on surfaces with retention of activity. For example, functional proteins and peptide epitopes modied with a ferrocene moiety have been immobilized on cucurbit [7]uril (CB7) surfaces with applications in protein arrays. [1][2][3] Similarly, beta-cyclodextrin (bCD) monolayers have been widely studied for the immobilization of ferrocene-labeled proteins or peptides via the ferrocene-bCD host-guest binding.…”

“…4,5 However, the relatively weak binding of bCD to ferrocene necessitates multivalent interactions to enable efficient surface immobilization on bCD monolayers. 6,7 Rapid and efficient supramolecular protein and cell adhesion thus requires new guest molecules with alternative chemotypes and strong binding affinities to bCD-functionalized surfaces.…”
