Hispidulin-7-O-neohesperidoside from Cirsium japonicum var. ussuriense attenuates the production of inflammatory mediators in LPS-induced raw 264.7 cells and HT-29 cells

Abstract: Background:Inflammatory bowel disease (IBD) is characterized by chronic inflammation of the gastrointestinal tract and involves secretion of inflammatory mediators. The flavone diglycoside hispidulin-7-O-neohesperidoside (HN) isolated from the methanolic extract of aerial parts of Cirsium japonicum var. ussuriense, but its pharmacologic activities, with the exception of alleviation of alcohol toxicity, have not been investigated to date. Objective:The aim of the present study was to investigate the anti-inflamm… Show more

“…TLR4 binding of LPS initiates the nuclear factor-kappa B (NF-κB) pathway leading to production of proinflammatory cytokines like IL-6 and IL-1β, resulting in fever and in worst case sepsis [16,17]. The murine macrophage cell line RAW264.7 has often been used for immunological studies of the gut [25,27,28]. It has also been used to test immunological effects of milk proteins and EVs [5,8,18].…”

“…Dinatin can inhibit the expression of HIF-1 α protein by activating AMPK pathway [ 52 ]. It can inhibit the production of inducible nitric oxide synthase (iNOS) protein and inflammatory cytokines such as TNF- α , IL-1, and IL-8 induced by lipopolysaccharide in Raw264.7 and human colon cancer (HT29) cells [ 84 ]. It can also be used to treat osteoporosis and prevent bone loss by activating AMPK signaling [ 85 ].…”
