Histamine-degrading enzymes as cellular markers of acute small bowel allograft rejection

Abstract: Intestinal histaminedegrading enzymes diamine oxidase (DAO) and histamine N-methyltransferase (HNMT) activities are relatively constant per individual and bowel segment, and they reflect the functional integrity of the intestinal mucosa. It was, therefore, hypothesised that a decrease in these enzymes could be indicative of acute rejection of an intestinal allograft. Enzymatic activities of DAO and HNMT were determined in mucosal biopsies of isogeneic (Lewis-toLewis, n = 48) and allogeneic (Brown Norway-to-Lew… Show more

“…The activity of histamine-degrading enzymes diamine oxidase and histamine Nmethyltransferase (HNMT) was found to decrease in the mucosa of rejecting rat allografts and revealed a strong negative correlation with the histological rejection score, whereas isografts showed rather constant tissue enzyme activity up to eight days after ITx [61]. These two enzymes are relatively constant within each individual and bowel segment and reflect the functional integrity of the intestinal mucosa.…”

“…It was difficult to determine which was the best strain to use to establish a useful AR model and the ideal moment to determine the typical histological features [67][68][69][70][71][72]. We initially used Wistar rats as recipients based on previous publications from our center (not directly related to rejection) [17,[19][20][21].…”
