Histamine Release from Washed Whole Blood

Abstract: Measurement of histamine release from basophilic leukocytes is a useful tool in the diagnosis of type I allergy. The modifications described in this paper – among others, the use of washed whole blood – offer several advantages over existing methods and render the procedure feasible as a routine diagnostic test.

“…a patient with exacerbation of asthma when withdrawn from mediation prior to BPT, HR was always carried out. The method used was a modified fluorimetric determination of histamine based on histamine-release from whole blood, and the method has been described in detail elsewhere (9). Estimations were performed by Allergologisk Laboratorium, Copenhagen.…”

“…The histamine release after one hour incubation at 37°C of blood cells mixed with a fixed amount of the sample used for the immunization was analyzed in accordance with the method described earlier. 50 The allergy response is expressed as the mean histamine release from blood cells of 10 rabbits. a possible connection with the propensity of insulin to fibrillate in the pump systems (see section 6).…”

“…Mould-induced histamine release was also performed from a serum-free cell suspension which has been found suitable for histamine release experiments as it avoids a significant loss of basophils during the isolation procedure (9). For that purpose, another 36 ml of heparinized peripheral blood were centrifuged at 350 X g for 10 min.…”
