“…This finding is consistent with several reports on expression of PDE4 mRNAs in these cells from rodents and humans (19,26,35). PDE4 inhibitors also have been used to indirectly assess the presence of PDE4 in neutrophils (3,26,36).…”

“…Postsynaptic H 2 receptors are known to be coupled to elevated AC activity (Lemos Legnazzi et al, 2000) and increase PDE4 activity 2-to 3-fold in blood mononuclear white cells (Delgado et al, 2003) and neutrophils (Dasi et al, 2000), explaining the increased radiotracer binding measured in our study. While radioactivity levels were significantly elevated in the heart following thioperamide injections, these increases were not blocked by (R)-rolipram, suggesting that changes in total binding are due to factors affecting non-specific binding of (R)-[ 11 C]rolipram.…”

“…Quantitative data of relative gene expression were determined by the comparative Ct method (⌬⌬Ct), as described by the manufacturer (PE-ABI PRISM 7700 sequence detection system) and previously reported. 18 Glyceraldehyde 3-phosphate dehydrogenase (GAPDH) was the endogenous control gene. TaqMan predevelopment assay reagents were used to determine IL-8 mRNA, and TaqMan RT reagents were used to generate cDNA.…”
