Histone Deacetylase 1 Down‐Regulation on Developmental Capability and Histone Acetylation in Bovine Oocytes and Parthenogenetic Embryos

Abstract: Histone deacetylase 1 (HDAC1) is one of the most conserved enzymes present in the nuclei of cells, including bovine oocytes and pre-implantation embryos. However, the biological functions of HDAC1 in supporting the growth and development of bovine oocytes and embryos are still not fully elucidates. In this study, three siRNAs (si299, si672, and si1272) targeting to HDAC1 mRNA sequence were designed. After transfection into bovine fibroblast cells, si299, the most effective one in HDAC1 knock-down, was selected… Show more

“…Oocyte collection and in vitro maturation were similar to previous research 50 . Briefly, bovine ovaries were collected from a local abattoir and transported to the laboratory within 4 h, then cumulus-oocyte complexes (COCs) were aspirated from the follicles using an 18-gauge needle attached to a 10 ml syringe.…”

“…It seems that high a level of histone acetylation was correlated with high developmental competence. However, knocking down histone deacetylase 1 (HDAC1) decreased the blastocyst formation rate of bovine parthenogenetic embryos despite the increase in acH3K14 [17]. The relationship between acetylation and developmental competence still needs to be further studied.…”

“…The mature oocytes were treated with 5 mM ionomycin for 5 min, followed by treatment with 2 mM 6-DMAP in synthetic oviduct fluid (Sofaa) [17] containing 10% FBS for 3.5 h at 38.5 C in 5% CO 2 and 95% humidified air. Treated oocytes were then washed in Sofaa three times and divided into two groups.…”

“…Then, COCs with intact, unexpanded cumulus cells were divided into two groups. One group of 20e25 COCs was cultured for 20e22 h at 38.5 C in 100 ml of maturation medium [17] in a humidified 5% CO 2 incubator. The second group was cultured in 100 ml of maturation medium supplemented with 200 mM RG108.…”
