Histopathological, immunohistochemical and biochemical alterations in liver tissue after fungicide-mancozeb exposures in Wistar albino rats

Abstract: To evaluate the histopathological, immunohistochemical, and biochemical effects of liver changes after mancozeb administration. Methods: Rats were divided into groups-the control group (n=7) and the mancozeb group (n=7)-, given 500 mg/kg mancozeb dissolved in corn oil daily for four weeks by an orogastric tube. Caspase-3 and tumor necrosis factor-alpha (TNF-α) primary antibodies were used for immunohistochemical analysis. Results: Serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) values … Show more

“…The current study of cell morphology levels found necrotic hepatitis, lobular inflammation, and hepatocellular ballooning after obtaining Mz, which were in accordance with earlier research that illustrated the focal necrosis of hepatocytes in Mz-treated rats [ 14 ]. The hepatotoxicity of Mz on histological features such as inflammatory infiltration and balloonization has also been reported [ 44 ].…”

“…Aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were altered in a concentration-dependent manner [ 13 ]. Mz also affected the histological features of the liver; 500 mg/kg of Mz induced hepatocyte degeneration, inflammation, and venous dilatation [ 14 ]. The antioxidant enzyme activity was reduced, which indicated increased lipid peroxidation (LPO) in rats exposed to Mz at doses of 250 and 500 mg/kg for 40 days [ 15 ].…”

“…Pes ticides are mainly used to des troy pes ts, living organisms with the same physiology and anatomy as other essential organisms like bees. These pes ticides target body sys tems like the nervous sys tem and other crucial sys tems in organisms, thereby disrupting and inhibiting the normal functioning of the physiology [5,6].Theextensive and careless application of chemical pes ticides on farms and agricultural products with little to zero knowledge of their proper application methods by mos t farmers brought about the unexpected. As a result, several synthetic chemical pes ticides throughout his tory have found their way into water bodiesthroughrunoff, windaction, andmanyother processes.…”

“…MNZ and ETU can cross the placental barrier, directly affecting reproduction at preconception, pregnancy, and birth stages [ 28 ]. A number of in vitro studies report that ETU reactivity is associated with direct DNA damage [ 29 , 30 , 31 ], nitrosamine formation [ 32 ], apoptosis induction [ 33 ], and the altered expression of genes involved in oxidative stress response [ 34 , 35 ], overall causing the gradual accumulation of damage to intracellular macromolecules exerted by reactive oxygen species (ROS) [ 36 , 37 ]. The consequent oxidative stress might be pivotal in the pathogenesis of thyroid, neural, and immune toxicity [ 38 ].…”
