Homocysteine aggravates DNA damage by impairing the FA/Brca1 Pathway in NE4C murine neural stem cells

Abstract: There is existing evidence that elevated homocysteine (Hcy) levels are risk factors for some neurodegenerative disorders. The pathogenesis of neurological diseases could be contributed to excessive cell dysfunction and death caused by defective DNA damage response (DDR) and accumulated DNA damage. Hcy is a neurotoxic amino acid and acts as a DNA damage inducer. However, it is not clear whether Hcy participates in the DDR. To investigate the effects of Hcy on DNA damage and the DDR, we employed mitomycin C (MMC… Show more

“…Sii‐Felice K et al discovered that transgenic mice with fanca −/− displayed increased SVZ cell apoptosis, chromosome instability, and increased depletion of NSPC pools, reflecting that FA/BRCA is critical for NSPC survival (Sii‐Felice et al, 2008b). Similar findings were made by Yan et al, where treatment of NE4C cells with homocysteine (Hcy), which can disrupt the FA/Brca1 pathway, was found to exacerbate the DDR induced by MMC in NSCs (Yan et al, 2020). Both studies demonstrate the importance of the Fanconi pathway for NSC in repairing DNA damage and combating ageing.…”

“…Growth media formulations are usually serum-free media used for primary NSC cultures obtained from mice or rats, composed of DMEM containing a 2% B27 supplement, 20 ng/mL EGF, 20 ng/mL bFGF, 2 µmol/mL L-glutamine, and 100 U/mL penicillin and streptomycin with or without 4 mg/L folic acid [ 14 , 34 ]. On the other hand, media used for immortalized cell lines usually contain animal sera, and they are formulated as follows: Dulbecco’s modified Eagle’s medium (5 mM glucose) supplemented with 10% fetal bovine serum, 5% horse serum, 2 mM glutamine, 100 U/mL penicillin, and 100 mg/mL streptomycin; or Minimum Essential Medium (MEM; Gibco, Carlsbad, CA, USA) with 10% fetal bovine serum, 2 mM glutamine, 100 U/mL penicillin, and 100 μg/mL streptomycin, for NE4C cells [ 32 , 35 ].…”
