HPLC method for amino acids profile in biological fluids and inborn metabolic disorders of aminoacidopathies

Abstract: Quantification of total and individual amino acids in biological fluids such as plasma, urine and cerebrospinal fluid has an important diagnostic implication in laboratory medicine. The present paper describes protocols for the assay of total amino acids by modified method based on dinitrophenyl and HPLC profile involving pre-column derivatization with o-pthalaldehyde (OPA) derivatization, respectively. The method, based on the alkylation of-SH groups prior to OPA derivatization of amino acids followed by reve… Show more

“…This finding has been reported in several experimental studies involving Fanconi syndrome induced by either maleate (Nissim and Weinberg, 1996) or ifosfamide (Mohrmann et al, 1993). An ingenuous method (Babu et al, 2002) was used for the identification of cysteine and tyrosine in the urine of carboplatin treated animals. Results showed amplified cysteinuria and tyrosinuria affording credence to our speculation that carboplatin might induce a Fanconi-like condition in rats.…”

“…Soluble sugars were extracted according to the modified method of Babu et al (2002). Biological material (400 mg) was ground in mortar with 2 mL of 80% (v/v) ethanol and centrifuge at 6000 g for 20 min.…”

“…Generally applied reagent for ARG derivatization, some of its metabolites and selected polyamines (PUT, SPERMD and SPERM) in human urine samples were OPA [38][39][40]42,[45][46][47]53,58,59]. Review articles about other reagents used for derivatization of different amino acids and various polyamines were presented by Teerlink [38], Molnár-Perl [60] and Fekkes [61].…”

“…GLN in urine samples was determined frequently simultaneously with ARG and some of its metabolites (e.g. CIT, ADMA, SDMA and ORN) [42,[45][46][47]. Several chromatographic methods have been published for quantification of PUT, SPERMD and SPERM in urine [48][49][50][51][52][53][54][55].…”
