Human Adenovirus from Subgenus D: Restriction Mapping of Types 9 and 19 and Characterization of a New Genome Type

Abstract: SUMMARYA strain of adenovirus (AV) was isolated from the stools of a child with acute gastroenteritis. This virus (strain 208) was specifically neutralized by antiserum to AVI9, a species belonging to subgenus D and previously reported to cause keratoconjunctivitis. However, restriction analysis of strain 208 showed it to be distinct from AV19 and the other 40 human AV serotypes presently known. When calculated from 18 restriction patterns, the proportion of comigrating fragments common to AV19 and strain 208 … Show more

“…The genetic relation to AV15, with only 51-63% comigrating fragments, is in the same range as that found between different prototypes of subgenus D [22]. It is noteworthy that the BamHX and Hind 111 maps of AV9, reported by Gomes and Niel [10], are in agreement with ours; these authors used similar meth ods to determine the DNA polarity. Con cerning the different location of polymor phic sites between AV9 and 15, 3 of the 12 sites were in the left and 9 in the right half of the genome.…”

“…Hence we established physical maps of AV9 and 15, and of the different genome types of AV15/H9. After all experiments were com pleted, Gomes and Niel [10] reported physi cal mapping of AV9 and 19 prototypes with several restriction enzymes; their Bam\\\ and ///«dill maps of AV9 corresponded to our results.…”

“…Furthermore, three restriction patterns of genome types of the intermediate strains AV15/H9 [3] correspond to the following prototypes which can, therefore, also be considered as physically mapped: HindlII pattern (2) =AV17, BgllI pattern (3)=AV10, and BamHI pattern (4)=AV10. Gomes and Niel [6] reported data on physical maps of AV9 and AV19 with HindlII and BamHI.…”

“…Until now, physical mapping has been done only with a few types and restriction enzymes [3,6,11] and Adrian et al (unpublished results on AV8). AV types of subgenus D show a high degree of DNA homology, determined by liquid hybridization [7].…”
