“…We exposed H295R cells to different concentrations of TBC in the absence or presence of 10 lM forskolin for 72 h, and then the concentrations of two steroid hormones, E2 and T, in the culture medium supernatant and the relative expression level of nine steroidogenic-related genes (StAR, 3bHSD2, CYP17, CYP21, CYP19, HMGR, 17bHSD1, 17bHSD4 and CYP11A) were measured. Our results showed that forskolin treatment of the H295R cells resulted in more hormone secretion and a generally increase expression of steroidogenic-related genes, which confirms its capability for inducing steroidogenesis in H295R cells and is consistent with previous reports (Gracia et al 2006;Hecker et al 2006). Additionally, TBC exposure caused significantly decrease in steroid hormones in H295R cells.…”

“…These include simazine, PCB126, BADGE, and TCDD (Hinson and Raven, 2006;Hecker et al, 2006;Villeneuve et al, 2007;Harvey et al, 2007). To examine the effects of these chemicals on POMC:EGFP expression in our transgenic fish as well as endogenous POMC expression in zebrafish, Tg (POMC: EGFP) reporter fish and wild-type zebrafish were treated with those chemicals for 4 days starting at 1 dpf (Table 1).…”

“…Upon cAMP stimulation, these cells produce all the adrenal steroid hormones found in the adult adrenal cortex (Gazdar et al, 1990;Sanderson, 2006). H295R cells have been employed for studying the biological effects and mechanisms of action of a variety of chemicals (e.g., Ding et al, 2007;Hecker et al, 2006Hecker et al, , 2007Hilscherova et al, 2004;Li and Wang, 2005;Sanderson, 2006;Zhang et al, 2005). In the present study, HPLC-MS/MS was used to examine the effects of 8:2 FTOH on the production of seven steroid hormones: progesterone, 17α-OH-progesterone, androstenedione, testosterone, deoxycorticosterone, corticosterone and cortisol.…”
