Human Cytomegalovirus Replication and Infection-Induced Syncytia Formation in Labial, Foreskin, and Fetal Lung Fibroblasts

Abstract: Only a handful of cell types, including fibroblasts, epithelial, and endothelial cells, can support human cytomegalovirus (CMV) replication in vitro, in striking contrast to the situation in vivo. While the susceptibility of epithelial and endothelial cells to CMV infection is strongly modulated by their anatomical site of origin, multiple CMV strains have been successfully isolated and propagated on fibroblasts derived from different organs. As oral mucosal cells are likely involved in CMV acquisition, we sou… Show more

“…The Jurkat cell line, which closely resembles human T lymphocytes, is widely utilized as a valuable tool in T cell research ( 43 ). In our study, we employed HFFs as infection cells because they are predominant in the pool of HCMV-infected cells in vivo and have been broadly used for propagation in cell culture ( 44 , 45 ). The experiments proved a significant reduction in viral growth in HCMV-infected HFFs that were co-cultured with CD3/CD28-activated Jurkat cells compared to HFFs cultured alone.…”

“…The fibroblasts produced viruses that could infect both fibroblasts and EC. HCMV replication in human fibroblast associated with different miRNA expressions has been established in several reference papers [63][64][65][66][67]. The enhanced synthesis of non-coding RNA molecules is almost certainly driven by a coupling effect between the viral infectious process and the host genomic response.…”
