Human fused NKG2D–IL-15 protein controls xenografted human gastric cancer through the recruitment and activation of NK cells

Abstract: Interleukin (IL)-15 plays an important role in natural killer (NK) and CD8+ T-cell proliferation and function and is more effective than IL-2 for tumor immunotherapy. The trans-presentation of IL-15 by neighboring cells is more effective for NK cell activation than its soluble IL-15. In this study, the fusion protein dsNKG2D-IL-15, which consisted of two identical extracellular domains of human NKG2D coupled to human IL-15 via a linker, was engineered in Escherichia coli. DsNKG2D-IL-15 could efficiently bind t… Show more

“…The safety of adoptive transfer of autologous or allogeneic NK cells has been proved; however, efficacy is limited by their poor proliferation and persistence in vivo and the increase in Tregs related to IL-2 administration (53). Administration of exogenous IL-15 has been shown to improve survival, in vivo persistence, and therapeutic efficacy without inducing the survival and expansion of Tregs (54,55). A human NKG2D-IL-15 fusion protein could efficiently bind to major histocompatibility complex class I polypeptide-related sequence + tumor cells and stimulated NK cell activity by transpresentation of IL-15, thus exhibiting higher control efficiency of the growth of xenografted human gastric cancer via the NK cell recruitment and activation (55).…”

“…IL-15 is a pleiotropic cytokine that belongs to the 4-a-helical bundle family of cytokines and acts via the attachment to the receptor complex comprising IL15Rα (CD215), IL2Rβ (CD122) and the common γ (CD132) chains [217]. IL-15 is involved in the control of development, homeostasis, and functions of T cells, NK cells and NK-T cells [218,219]. It is also a vital cytokine for the control of B cells, DCs, macrophages, and mast cell functions.…”
