Human glioblastoma cells produce 77 amino acid interleukin-8 (IL-877)

Abstract: The production of interleukin 8 (IL-8), a neutrophil chemotactic factor, and its amino acid sequence were examined in glioblastoma cell lines in vitro. Neutrophil chemotactic activity was demonstrated in 9 conditioned media of 15 human glioblastoma cell lines. Tumor necrosis factor (TNF)-alpha stimulated secretion of the activity in 7 lines and induced secretion in 4 other lines. ELISA quantification disclosed that the conditioned media contained interleukin 8 (IL-8) in an amount equivalent to the chemotactic … Show more

“…These results are consistent with previous studies showing that endothelial cells derived from ovarian and kidney tumors constitutively produce high levels of IL-8 (25). To eliminate the possibility that contaminating glioma cells are responsible for this IL-8 production (5,26,27), extensive analyses were done to confirm that TuBECs were exclusively endothelial cells (19).…”

“…Our previous work (C.D., M.R.J., unpublished data, May 2001) failed to detect any colony-stimulating factor (eg, granulocyte (G)-CSF or granulocyte-macrophage (GM)-CSF) made by T9 or T9-C2 cells in vitro that could account for the presence of the granulocytes in vivo. This prompted us to examine possible chemokines, because human gliomas 22 and rat C6 gliomas 23 produce IL-8 and the rodent IL-8 equivalent known as CINC, respectively. By RT-PCR, both T9 and T9-C2 cells produced CINC-specific mRNA (Figure 3).…”

“…HIV), malignant brain neoplasm (Tada et al, 1993), multiple sclerosis (Spuler et al, 1996), and neurosurgical procedures (Woiciechowsky et al, 1997) are accompanied by a significant inflammatory response in the brain. This inflammatory reaction is usually characterized by microcirculatory disorders (Müller et al, 2001), edema formation, or swelling of the brain parenchyma.…”
