“…Using the rat T9 tumor model, we have seen that both PMNs and macrophages contributed to the paraptotic killing of the mM-CSF-expressing T9-C2 cells. 31 However in this study, we have only observed a monocytic/macrophagic infiltrate within the implanted MG-2F11 cells. Thus, human tumor cells can be killed via paraptosis by only monocytes/macrophages.…”

“…Although initially disputed, several forms of caspase-independent apoptosis [33][34][35] or forms that are similar to apoptosis ('paraptosis') but also do not involve caspases have been reported. 30,36 Sperandio and colleagues (2000) characterized 'paraptosis' in 293T cells and mouse embryonic fibroblasts as a form of cell death without nuclear fragmentation and apoptotic bodies, but with cytoplasmic vacuolation and late mitochondrial swelling. In addition, death was independent of caspase activity, Bcl-X L , and was executed without DNA fragmentation.…”

“…57,58 In vivo there is evidence that inducible nitric oxide synthase and peroxynitrite are present at the subcutaneous mM-CSF tumor cell rejection sites. 16,37 In the current study the exact macrophage-derived mediator responsible for paraptosis induction was not addressed. ROS were detected within 18 min and exogenously added H 2 O 2 and arachidonic acid (data not shown) caused cellular swelling and vacuolization of the U251 glioma cells.…”

“…The in vivo morphology of the destruction of the rat and human glioma cells was closely examined. 16,37 After subcutaneous injection, membrane M-CSF transduced gliomas began dying within 4 h of the development of the swelling and vacuolization process of paraptosis. In the current study, a putative molecular mechanism was in- (Figure 3).…”
