Human IGF1 pro-forms induce breast cancer cell proliferation via the IGF1 receptor

Abstract: Our data indicate that IGF1 pro-forms can induce breast cancer cell proliferation via the IGF1R, independent from the mature IGF1 form. These results underline the importance of an accurate assessment of the presence of IGF1 pro-forms within the breast cancer microenvironment.

“…ProIGFs activate either the MAPK or the PI3K/AKT arm of IGF-1R downstream signaling in a cell type-dependent manner. Contrary to transgenic mice overexpressing IGF-1Ea, AKT phosphorylation is affected by glycosylated pro-IGF-1Ea in MCF-7 human breast (adenocarcinoma) epithelial cells [27,29]. However, among proIGFs, we cannot exclude the existence of distinct downstream signaling pathways that are divergent from that of mature IGF-1.…”

“…Intracellular IGF-1 seems to be mainly expressed as proIGF-1 that is extracellularly converted into mature IGF-1 [26,27]. Although IGF-1’s biological activity is mainly exerted by the mature peptide, recent evidence suggests that the various prohormones exert functional roles [28].…”

“…Recent investigations have stressed the role of circulating growth factors, prohormones, and sexual hormones, which are able to stimulate cancer cell proliferation through the activation of their respective receptors [19,20]. In particular, the phosphoinositide 3-kinase-AKT-mammalian target of rapamycin (PI3K-AKT-mTOR) pathway is a frequently hyperactivated pathway in cancer and is important for tumor cell growth and survival.…”
