Hydrogen Sulfide Gas Exposure Induces Necroptosis and Promotes Inflammation through the MAPK/NF-κB Pathway in Broiler Spleen

Abstract: Hydrogen sulfide (H2S) is one of the main pollutants in the atmosphere, which is a serious threat to human health. The decomposition of sulfur-containing organics in chicken houses could produce a large amount of H2S, thereby damaging poultry health. In this study, one-day-old broilers were selected and exposed to 4 or 20 ppm of H2S gas (0-3 weeks: 4±0.5 ppm, 4-6 weeks: 20±0.5 ppm). The spleen samples were collected immediately after the chickens were euthanized at 2, 4, and 6 weeks. The histopathological and … Show more

“…Given the impressive results observed in AGS cell line by PRU treatment, we were curious to determine the mechanism of its action or pathway leading to necroptosis cell death. The mitogen-activated protein kinase (MAPK) pathway is an important signaling pathway involved in oxidative stress, necroptosis, inflammation, and various pathological mechanisms [ 40 ]. In this aspect, we studied the expression levels of mitogen-activated protein kinase (MAPK) family of proteins JNK, p38, ERK, and its phosphorylated active forms—p-JNK, p-p38, and p-ERK—by western blot analysis.…”

“…Furthermore, H 2 S exposure (0–3 weeks: 4 ppm, 4–6 weeks: 20 ppm of H 2 S gas) was shown to induce oxidative stress and energy metabolism dysfunction. It also led to necroptosis, activated the MAPK pathway, and triggered the NF-κB pathway associated with a promotion of inflammatory response in chicken spleens [ 251 ]. To study the immunotoxicity of H 2 S, 1 day old broiler chicks were exposed to atmospheric H 2 S for 42 days.…”

“…NF-kB is a vital pathway for regulation of the release of inflammation factors. MAPK is a positive regulator of the NF-kB signaling pathway [30]. Furthermore, NF-kB was triggered by NOD2, which was reported to promote the innate inflammatory response by activating NF-kB signaling.…”
