Hydrophilic Chlorin e6-Poly(amidoamine) Dendrimer Nanoconjugates for Enhanced Photodynamic Therapy

Abstract: In photodynamic therapy (PDT), chlorin e6 (Ce6), with its high phototoxic potential and strong absorption of visible light, penetrates deeply into photodamaged tissue. However, despite this fact, the direct application of Ce6 to PDT has been limited by its low water solubility and poor cancer cell localization. To ameliorate this situation, we report herein on the use of a hydrophilic nanoconjugate (DC) comprised of Ce6 and poly(amidoamine) dendrimer, which improves the water solubility and intracellular uptak… Show more

“…Therefore, to verify the enhanced bioactivity of the CEPP20 monolith, 1 O2 generation from the CEPP monoliths during laser irradiation was quantitatively determined using 9,10-dimethylanthracene (DMA) as the 1 O2 chemical probe. Fluorescent DMA reacts selectively with the 1 O2 to form endoperoxide, causing a reduction in the fluorescence of DMA [25]. The 1 O2 formation from the CEPP monoliths increased with increasing immobilized PpIX content on the monolith surface.…”

“…[ 227 ] In another work, PAMAM dendrimers conjugated to Ce6 were also shown to be potent nanophotosensitizers in vitro against human cervical cancer cells, inducing photoactivated apoptosis in a more effective way than Ce6 alone. [ 228 ]…”

“…To overcome the drawbacks of poor water solubility and undesirable cell localization of Ce6, Lee et al designed a novel hydrophilic nano‐copolymer (DC) with poly‐amidoamine dendrimers. The spherical DC with an average particle size of 61.7 nm displayed not only better cellular uptake but also stronger cytotoxicity than Ce6 alone (Lee & Kim, 2018 ).…”
