Actinobacillus pleuropneumoniae Osteomyelitis in Pigs Demonstrated by Fluorescent In Situ Hybridization

Abstract: Abstract. Necrotizing osteomyelitis and fibrinopurulent arthritis with isolation of Actinobacillus pleuropneumoniae serotype 2 is reported in two pigs from a herd with lameness and mild coughing problems among 8 to 12-week-old pigs. Application of fluorescent in situ hybridization targeting 16S ribosomal RNA of A. pleuropneumoniae in formalin-fixed tissue was performed to verify the association of A. pleuropneumoniae with the bone and joint lesions. By in situ hybridization A. pleuropneumoniae was demonstrated… Show more

“…For the specific detection of A. pleuropneumoniae, sections were examined by FISH using a fluorescein isothiocyanate (FITC)-labelled oligonucleotide probe (Ap185; MWGbiotech AG, Ebersberg, Germany), with the sequence 5 0 -CCCACCCTTTAATCCATAG-3 0 , targeting 16S rRNA of A. pleuropneumoniae, as previously described. [29][30][31] In each test round, sections from lung tissue containing A. pleuropneumoniae, as verified by bacteriology and FISH, were included as positive staining controls.…”

“…FISH has been used to visualize the spatial distribution of Escherichia coli in intestinal mucosa (42); to evaluate colonization of mouse intestines by Salmonella enterica serovar Typhimurium (32); and to specifically detect Salmonella serovars in pig intestines and mouse lungs (40), Actinobacillus pleuropneumoniae strains in diseased porcine tissues (23,26), Brachyspira (Serpulina) pilosicoli infections in the intestines of growing pigs (11,27), Lawsonia intracellularis in porcine proliferative enteropathy (10), Streptococcus suis infection in pigs (12), and bacteria in blood cultures (25,29).…”

“…The results of an earlier study indicate that fluorescent in situ hybridization using a DNA probe designed from the 16S ribosomal RNA of A. pleuropneumoniae can detect the 12 serotypes of A. pleuropneumoniae [9,10]. However, that study did not examine lung tissue from pigs infected with all of the A. pleuropneumoniae serotypes, and the use of fluorescent in situ hybridization did not preserve cellular and histological details.…”
