Gbx2 regulates thalamocortical axon guidance by modifying the LIM and Robo codes

Abstract: SUMMARYCombinatorial expression of transcription factors forms transcriptional codes to confer neuronal identities and connectivity. However, how these intrinsic factors orchestrate the spatiotemporal expression of guidance molecules to dictate the responsiveness of axons to guidance cues is less understood. Thalamocortical axons (TCAs) represent the major input to the neocortex and modulate cognitive functions, consciousness and alertness. TCAs travel a long distance and make multiple target choices en route … Show more

“…Taking advantage of the mosaic nature of creER-mediated recombination induced by tamoxifen, we deleted Gbx2 in a mosaic manner in the thalamus of Gbx2 creER/F embryos as described previously (Chen et al, 2009; Chatterjee et al, 2012). As Gbx2 acts cell autonomously to regulate thalamocortical axon guidance, all mosaic deletion embryos were verified by abnormal thalamic axon trajectories examined by GFP fluorescence that was expressed from the Gbx2 cerER locus as described previously (Chatterjee et al, 2012).…”

“…Recently, LHX9 was also found to be expressed in a subset of hypocretin neurons in the hypothalamus and to have a crucial role in regulating somnolence (Dalal et al, 2013). Further, combinatorial expression of LIM-HD factors LHX9 and LHX2 has been demonstrated during the development of several systems (Nakagawa and O’Leary 2001, Wilson et al 2008, Abellan et al 2009, Peukert et al 2011, Chatterjee et al 2012). …”
