Herpesvirus saimiri Transformation of Human T Lymphocytes

Abstract: Viral transformation of T cells is an effective method for obtaining large numbers of T cells that are easily maintained in the laboratory. This unit describes a method for generating antigen-independent, virally-transformed T cells using a T-lymphotropic primate gamma-2 herpesvirus, Herpesvirus saimiri (HVS; strain C488). Support protocols for preparing and titrating HSV C488 stocks and testing the functional status of transformed T cells are also included.

“…After 24-h culture, 10 ng/ml rIL-2 (Thermo Fisher Scientific) was added to the medium. Subsequently, medium and IL-2 were renewed every 48 h. PBMCs were transformed with H. saimiri strain C488, as previously described, to ensure continuous growth (Fleckenstein and Ensser, 2004). Saimiri-transformed cells were cultivated in Panserin/RPMI 1640 (ratio 1:1) supplemented with 10% FBS, l-glutamine, gentamycin (1×), and 10 ng/ml human rIL-2 (Thermo Fisher Scientific).…”

“…However, co-species transmission to New World non-human primates can lead to the development of acute T-cell lymphomas (Fickenscher and Fleckenstein, 2001). Interestingly, SaHV-2 can induce transformation of human and rabbit T lymphocytes (Fleckenstein and Ensser, 2004). The transforming capability of SaHV-2 has been identified to be mainly driven by two viral proteins termed Stp and Tip (Fleckenstein and Ensser, 2007).…”
