Introduction: The aim of this study was to evaluate the antibacterial efficacy of Vitex negundo Linn. extract as root canal irrigant against Enterococcus faecalis and its penetration into root dentin. Methods and Materials: Forty single rooted premolars were randomly divided into 4 groups: 3% Sodium hypochlorite (NaOCl), 2% Chlorhexidine (CHX) , 100mg/ml Vitex negundo Linn. and saline as control all mixed with Rhodamine B dye. Test samples were analysed for bacterial count before and after irrigation using absorbent paper points and the colony forming units were recorded and measured. Sectioning of the samples was performed at three levels 3mm,6mm,9mm from apex and then these samples were analysed using confocal laser scanning microscopy for penetration depth of the irrigant within the dentinal tubules. Paired t-test and ANOVA test were used to perform statistical analysis with level of significance set at 0.05 Results: The mean CFU/ml count of Enterococcus facealis reduced significantly in all the groups post irrigation. All the irrigants showed maximum penetration depth at coronal third level compared to middle and apical third level respectively. The penetration depth of NaOCl group was better when compared to CHX group and Vitex negundo Linn. group but the difference was statistically not significant. Conclusion: Although 3% NaOCl was the most effective irrigant, all agents exerted acceptable antimicrobial activity against Enterococcus faecalis and penetration depth within tubules of dentin.

The main aim of Endodontics is to either prevent the endodontic disease or to treat it. This is achieved by maintaining the pulp health in cases of pulp inflammation. If there is pulp necrosis, pulp health can be maintained by regenerating the healthy pulp tissue. Hence various researches and the clinical studies tried to regenerate the healthy functioning pulp-dentin complex. Regenerative dentistry aims to biologically replace the dental tissues along with their supporting structures. Regenerative Endodontics can be defined as “ biologically based procedures designed to replace damaged structures such as dentin, root structures, and cells of the pulp-dentin complex.” Root canal anatomy is complex, hence even after mechanical instrumentation, there exists some portions of the root canal that remain untouched. Only instrumentation cannot completely eliminate the microbes existing into the root canal. Hence there is need to additionally irrigate the canals and also using medicaments is required to disinfect the root canal and promote healing. This article aims at focussing on various antibiotics used and role of these materials to disinfect the canals during revascularization treatment.

Complete removal of infection, rendering root canal free of an organism is the prime aim of endodontic treatment. It cannot be achieved alone by biomechanical preparation due to the complexity of root canal treatment. Therefore the use of intracanal medicament is a must. The antibiotic paste is frequently used intracanal medicament, usually in regenerative cases. Hence the aim of the is to compare different irrigation technique for removal of double antibiotic paste from root canal through a stereomicroscope. 36 single-rooted were standardized to root length of 12mm then instrumented by Protaper rotary file up to size F4. Irrigation was done using sodium hypochlorite in between instrument change. The root canal was dried using paper point and filled by DAP and then randomly allocated to 3 groups(n=12) according to irrigation system used: conventional syringe irrigation (CSI), Passive ultrasonic irrigation(PUI), EndoVac (EV). Roots were then longitudinally sectioned using the diamond disk and studied under a stereomicroscope using a scoring scale. Data were evaluated using one way ANOVA and Tukey test. Among all experimental group CSI was least efficient. PUI and EV showed greater efficiency but no significant difference between PUI and EV, but a significant difference between CSI and PUI, EV (p< 0.05). Use of irrigation activation system results in efficient removal of DAP compared to CSI.
