In Vitro Evaluation of Proliferation and Migration Behaviour of Human Bone Marrow-Derived Mesenchymal Stem Cells in Presence of Platelet-Rich Plasma

Abstract: Objective. To access the effects of platelet-rich plasma (PRP) on the behaviour of human bone marrow-derived mesenchymal stem cells (hBMSCs), including proliferation and migration. Methods. PRP was diluted with DMEM/F12, resulting in concentrations of 1%, 2%, and 5%. The proliferation of hBMSCs was examined by 2 methods: cell-number counting with the haemocytometer method and the colony-forming unit-fibroblast (CFU-F) assay. Cell migration was evaluated using the scratch wound healing (SWH) assay; after that, … Show more

“…As an alternative to these approaches, the use of PRP as clinical-grade adjuvant to enhance the therapeutic effectiveness of engrafted MSC has been suggested by several studies highlighting that PRP treatment improves the angiogenic potential of MSC both in vitro and in vivo [29, 30] and stimulates the proliferation of MSC in vitro [31, 32]. However, systematic studies on whether PRP alters the repair properties of engrafted MSC in skin wound healing are lacking.…”

“…Only 6 of these 21 studies had a sample size of n ≥ 10 for both components (PPs and BM-MSCs) (Ben Azouna et al, 2012;Jenhani et al, 2011;Lucarelli et al, 2003;Moisley et al, 2019;Verrier et al, 2010;Yin et al, 2016); with the remaining having a small sample size of n ≤ 6 for one or both components. Discrepancy in sample size between PPs and BM-MSCs within the same study was also observed (Amable et al, 2014;Ben Azouna et al, 2012;Bernardi et al, 2017;Bernardi et al, 2013;Goedecke et al, 2011;Gottipamula et al, 2012;Infante et al, 2017;Jenhani et al, 2011;Kasten et al, 2008;Parsons et al, 2008;Perut et al, 2013;Prins et al, 2009;Vogel et al, 2006;Yin et al, 2016) (Table 2a,b and 3a,b).…”

“…The effect of incubation time on PPs was only assessed by 1 study (Parsons et al, 2008), whereby BM-MSCs incubated with 2.5 % PRC showed more proliferation in the first 24 h only, with proliferation equal to that of the control group at 48 h and inferior to control group at 72 h.…”
