MMP8polymorphism is associated with susceptibility to osteonecrosis of the femoral head in a Chinese Han population

Abstract: Osteonecrosis of the femoral head (ONFH) is an orthopedic refractory disease that adversely affects quality of life. Matrix metalloproteinase-8 (MMP-8) produced by the bone marrow has been implicated in the degradation of collagen during bone development. We assessed whether MMP8 polymorphisms are associated with ONFH. In a case-control study, using χ2 tests and genetic model analyses, we genotyped 5 MMP8 single-nucleotide polymorphisms (SNPs) in 585 ONFH patients and 507 healthy control subjects in a Chinese … Show more

“…In the present study, we investigated nine SNPs in the MMP8 and MMP9 genes. Among these SNPs, rs3740938, rs2012390, rs11225394, rs2274755 and rs3918254 were identified that have been associated with risk of osteonecrosis of the femoral head in the Chinese Han population 20 , 21 , 30 . We report for the first time that rs11225394 was associated with BC risk.…”

“…[15,16] MMP-8 was found to be correlated with RA and osteonecrosis of the femoral head. [17,18] Mattey et al found that bath AS disease activity index (BASDAI) was correlated with a group of clustered biomarkers consisting of MMP-8, MMP-9, hepatocyte growth factor, the chemokine, and CXCL8. However, there was no study revealed the association between MMP-8 and AS.…”

“…Although the suspected causes of osteonecrosis include ischaemia, MRI of our case revealed hypervascularity, presumably due to the newly formed feeder arteries from the primary cSCC lesion. In contrast, many kinds of factors, such as matrix metalloproteases (MMPs), transforming growth factor beta (TGF-β), interleukin (IL)-1, tumour necrosis factor alpha (TNF-α), and interferon alpha (IFN-α), are suggested to be involved in osteonecrosis through various effects, including inhibition of osteoblast differentiation and proliferation and promotion of bone cell death (10,11). Most of these factors could be produced in the tumour environment (12).…”
