Retracted: Addition of a single methyl group to a small molecule sodium channel inhibitor introduces a new mode of gating modulation

Abstract: Background and Purpose Aryl sulfonamide Nav1.3 or Nav1.7 voltage‐gated sodium (Nav) channel inhibitors interact with the Domain 4 voltage sensor domain (D4 VSD). During studies to better understand the structure‐activity relationship of this interaction, an additional mode of channel modulation, specifically slowing of inactivation, was revealed by addition of a single methyl moiety. The objective of the current study was to determine if these different modulatory effects are mediated by the same or distinct i… Show more

“…The binding site of TEL to Na + channels is likely at around ser571. 37 Reminiscent of a current study, 38 whether the methyl group at position 4 of the benzimidazole core embedded in the TEL molecule, which contributes to its PPARγ activity, is a potentially important moiety for its stimulatory effect on I Na remains to be studied extensively.…”

“…Therefore, the local anesthesia effect of benzoate ester was further conrmed. [45][46][47] Aer determining the local anesthesia effect of benzoate ester, the structure of the parent nucleus of cocaine (ecgonine) was modied and simplied. When the N-methyl of ecgonine was removed, there was no signicant effect on physiological activities, but the toxicity increased.…”
