Identification and Characterization of Optimal Gene Expression Markers for Detection of Breast Cancer Metastasis

Abstract: Sentinel lymph node (SLN) status is highly predictive of overall axillary lymph node involvement in breast cancer. Historically, SLN-positive patients have undergone axillary lymph node dissection in a second surgery. Intraoperative SLN analysis could reduce the cost and complications of a second surgery; however, existing histopathological methods lack standardization and exhibit poor sensitivity. Rapid molecular methods may lead to improved intraoperative diagnosis of SLN metastasis. In this study, we used a… Show more

“…Taken together with the correlation of GABRP with the BLBC subtype, metastatic spread to the brain, overall patient survival, and the migratory ability of BLBC cells described herein, GABRP may be a viable therapeutic target for treating BLBC. Supporting this possibility, GABRP is consistently detected in circulating breast tumor cells (51)(52)(53)(54) and in lymph nodes isolated from breast cancer patients (55). Targeting GABRP should be achievable because it is a cell surface receptor subunit.…”

“…As reported previously [17], in cases for which the BLN Assay result and the OHR were different, residual node homogenate or RNA extract from the assay testing was evaluated with other molecular markers for nodal metastases including B305D, B726, prolactin inducible protein, and prostate-derived Ets transcription factor [18], [19] and [20]. This testing was performed to confirm the BLN Assay results on the same tissue.…”

“…Not surprisingly, all top candidates have been identified in previous studies and well described in the literature. [23][24][25][26][27][28][29][30][31] A special approach was used to identify pancreasspecific markers. Pancreatic ductal adenocarcinoma develops from ductal epithelial cells that compose only a small percentage of all pancreatic cells (with acinar and islet cells composing the majority) in the normal pancreas.…”

“…24 A total of 23 tissue-specific marker candidates were selected for further RT-PCR interrogation on 205 metastatic carcinoma FFPE tissues by qRT-PCR. Of 23 tested markers, 13 were rejected based on their cross-reactivity, low expression level in the corresponding metastatic tissues, or redundancy.…”
