“…Analysis of the temporal kinetics of mda-5 expression indicate potentially novel and interesting features when compared to other IFN-dependent differentiation-induced genes isolated in our subtraction hybridization screens Huang et al, 1999b;Kang et al, 2001;Leszczyniecka et al, 2002). Biphasic induction of mda-5 by IFN-b and poly [I] : poly[C] was evident at the mRNA level.…”

“…It should be noted that treatment of HO-1 cells with IFN-b alone inhibits cell proliferation and increases melanogenesis, while combined treatment with MEZ induces changes in cell growth characteristics and morphology resembling a terminally differentiated state Jiang et al, , 1994. To define the molecular aspects of the terminal differentiation process, changes in gene expression patterns were analysed between undifferentiated and IFN-b þ MEZ differentiated cells by subtracted cDNA library screening approaches Jiang et al, 1994), including rapid subtraction hybridization (RaSH) (Jiang et al, 2000) and differentiation induction subtraction hybridization (DISH) (Huang et al, 1999b). Melanoma differentiation associated gene-5 (mda-5) was identified as an upregulated sequence in a cDNA subtraction library screening of terminally differentiating HO-1 cells by combined treatment with IFN-b and MEZ, although treatment with IFN-b is also sufficient to induce significant gene expression over basal levels Kang et al, 2002).…”

“…Due to their differential expression during melanocytic tumor progression, several genes or gene products have been recognized using various methods at the DNA, mRNA, and protein levels (Van Groningen et al, 1995;Hashimoto et al, 1996;Ishiguro et al, 1996;Simon et al, 1996;Duncan et al, 1998;Huang et al, 1999;Zendman et al, 1999;Habeck, 2001).…”

“…In human melanoma cells, treatment with the combination of fibroblast interferon (IFN-␤) ϩ mezerein (MEZ) results in irreversible growth arrest, loss of tumorigenic properties, and terminal cell differentiation (21,22). These changes in melanoma physiology involve specific temporally regulated modifications in gene expression (22)(23)(24). Potentially relevant genes associated with this process have been identified from a temporally spaced subtracted IFN-␤ ϩ MEZ-treated HO-1 human melanoma cDNA library by using several molecular approaches, including random screening of cDNAs, high throughput microchip cDNA microarrays and random cDNA clonal analysis, and screening by reverse Northern hybridization (8,23,24).…”

These approaches provide a molecular snapshot of genes regulated during terminal differentiation, and they have produced a useful database for determining the utility and efficacy of the RaSH approach.…”
