Identification and Validation of a Novel Prognosis Prediction Model in Adrenocortical Carcinoma by Integrative Bioinformatics Analysis, Statistics, and Machine Learning

Abstract: Adrenocortical carcinoma (ACC) is a rare malignancy with poor prognosis. Thus, we aimed to establish a potential gene model for prognosis prediction of patients with ACC. First, weighted gene co-expression network (WGCNA) was constructed to screen two key modules (blue: P = 5e-05, R^2 = 0.65; red: P = 4e-06, R^2 = −0.71). Second, 93 survival-associated genes were identified. Third, 11 potential prognosis models were constructed, and two models were further selected. Survival analysis, receiver operating charac… Show more

“…In line with this result, Fojo et al did not find significant differences in driver genes mutation and expression profiles between primary and metastatic ACCs [ 68 ▪ ]. This result is in partial contrast with a previous report by Gara et al who found a higher mutation rate in metastatic vs primary ACCs and an overlap of 37–57% in mutated genes among different sites from the same patient [ 69 ].…”

“…The identi cation of KIF2C as a noteworthy element in a newly developed prognostic prediction model for ACC underscores its potential involvement in determining the disease's outcome (70). The role of Matrix Metalloproteinase-9 (MMP-9) has been the subject of wide research in diverse cancer types, and its correlation with tumor advancement and metastasis has been established in numerous malignancies (71).…”

“…These nodes received training feedback via gene scores and weights and transmitted diagnostic results (predicting sample grouping) to the output layer (the third layer) 43 . Finally, we evaluated the ANN model's accuracy, and an ROC curve was plotted through the R package pROC (Xavier Robin [aut, cre], https://cran.r-project.org/web/packages/pROC/) to visualize the outcome 44 …”

“…43 Finally, we evaluated the ANN model's accuracy, and an ROC curve was plotted through the R package pROC (Xavier Robin [aut, cre], https://cran.r-project.org/web/packages/pROC/) to visualize the outcome. 44 2.8 | CDEG gene scores in test set samples R software was used to enter the CDEG expression matrix of the test sets (datasets GSE16088 and GSE14359) and its corresponding logFC values. The median expression level of CDEGs was then compared with their relative expression levels.…”
