Identification of a New Naphthalene and Its Derivatives from the Bulb of Eleutherine americana with Inhibitory Activity on Lipopolysaccharide-Induced Nitric Oxide Production

Abstract: A new naphthoquinone, (؊)-3-[2-(acetyloxy)propyl]-2-hydroxy-8-methoxy-1,4-naphthoquinone (1) was isolated from the bulb of Eleutherine americana MERR. et HEYNE (Iridaceae) together with two known compounds, eleutherinol (6) and 1,5-dihydroxy-3-methylanthraquinone (7) which were found in this species for the first time. The other known compounds, (؊)-isoeleutherin (2), (؉)-eleutherin (3), (؊)-hongconin (4), and (؉)-dihydroeleutherinol (5) which were reported previously from this species, were also isolated in t… Show more

“…When evaluated in vitro , they showed satisfactory action against inflammation induced by LPS in mouse macrophage RAW 264.7 cell lines. [85] Similarly, luteolin is a newer flavone which occurs indigenously in several plants and helps a lot for its antioxidant, antinflammatory and antiallergic functions as compared to other flavonoids. [86]…”

“…Although naphthoquinones have also a toxicological potential, mainly due to their ability to induce oxidative stress [8], they can be good lead compounds for new anti-inflammatory drugs, as several previous studies evidence. Bisnaphthoquinones inhibit platelet aggregation [9], plumbagin inhibits cytokines release [10], shikonin derivatives inhibit cyclooxygenase-2 expression [11], mast cell degranulation [12] and iNOS (an inducible calcium-independent isoform of nitric oxide (NO) synthases) [13], interfering with NO production [14]. However more studies are needed to confirm a really beneficial property.…”

“…Some condensed tannins were reported to inhibit melanin formation in melanocytes such as proanthocyanidins and procyanidin [22][23][24]. E. palmifolia contains several naphthalenes, namely eleutherol, eleutherin, and isoeleuthrin [25], and some naphthalenes have been reported to act as inhibitors of lipopolysaccharideinduced nitric oxide production [26]. Moreover, the freeradical scavenging activity of this extract may be related in part to the inhibition of melanin formation.…”
