Identification of biomarkers associated with partial epithelial to mesenchymal transition in the secretome of slug over-expressing hepatocellular carcinoma cells

Abstract: From our data we conclude that the exosomal proteins identified may be considered as potential non-invasive biomarkers for chemo-resistance and partial EMT in HCC.

“…Complete EMT is observed in individuals with vital role of it in metastasis initiation. Karaosmanoğlu Oand their group demonstrated reduction of E-cadherin and upregulation of ZEB2 in complete EMT, and meanwhile partial EMT involves increase of E-cadherin and decrease of vimentin and ZEB2 [87]. It is worth notifying that the EMT might causean increasein the cancer stem-like cells (CSCs), which refers to the highly tumorigenic subpopulation of tumor cells that exists at the top of the hierarchical tumor cell society [88,89], leading to tumor heterogeneity and therapeutic resistance [89].…”

“…HCC represents > 90% of primary liver cancers and is a major health problem due to its poor prognosis [8,20]. A number of risk factors for the development of HCC has been identified, and over 50% of HCC cases are attributable to persistent HBV The data shown are from one of three independent experiments which yielded similar results.…”

“…However, those biomarkers display limited sensitivity and specificity, especially with respect to early HCC stages and, therefore, combinations with other newly-identified candidate biomarkers are currently being evaluated [6]. Over the past years, technologies including whole genome sequencing (WGS), RNA-sequencing (RNA-seq) and proteomic profiling have led to a new era in biomarker development that has improved our understanding of complex interactions between proteins, genes and noncoding RNAs in hepatocarcinogenesis in different settings [7][8][9]. Most studies did, however, not distinguish HBV-related HCCs from HCCs resulting from other factors, and the analyzed tumor tissues were rarely matched with adjacent peritumor tissues (APTs) from the same patients.…”
