Identification of cytokine-induced neutrophil chemoattractants (CINC), rat GRO/CINC-2α and CINC-2 β, produced by granulation tissue in culture: purification, complete amino acid sequences and characterization

Abstract: Four basic neutrophil chemotactic factors (chemokines) have been purified from conditioned medium of granulation tissue obtained from carrageenin-induced inflammation in the rat. On the basis of their N-terminal amino acid sequences, one of the chemokines was identical with rat GRO/cytokine-induced neutrophil chemoattractant (CINC) which we reported previously, and another was identical with rat macrophage inflammatory protein-2 (MIP-2). Two other chemokines were novel chemoattractants related to MIP-2. The no… Show more

“…The cytokines with significant changes are involved in multiple cellular functions. For example, CINC-2a/b, CINC-3 and thymus chemokine play important roles in neutrophil recruitment (Nakagawa et al, 1994). Fratalkine mediates leukocyte adhesion (Jones et al, 2010) and IL-10 is an anti-inflammatory cytokine (Braat et al, 2006).…”

“…However, our study is the first to demonstrate the effect of brain death on the rat chemokines CINC-1 and CINC-3 and neutrophil integrins. CINC-1 is an 8-kD chemokine, structurally similar to human GRO and with properties similar to human IL-8 (28,29). CINC-3, a chemokine synonymous to rat macrophage inflammatory protein-2 (MIP-2), is strongly chemotactic for neutrophils and known to elicit a neutrophilic inflammatory response (30).…”

“…As members of the CXC chemokine family, rat CINC-1, CINC-2␣, CINC-2␤, and CINC-3 are neutrophil-specific chemokines that bind to a common receptor (CXCR2), show structural and functional homology to human IL-8, and are potent neutrophil chemoattractants in vitro and in vivo. [23][24][25][26][27] In a variety of acute vascular injury models, including a rat stroke model of middle cerebral artery occlusion with reperfusion and a rat liver transplant model with prolonged cold ischemia, upregulation of CINC has been demonstrated and correlated with neutrophil infiltration. 28,29 These acute-injury responses may play important roles in many pathological conditions, including acute coronary syndromes, as demonstrated by experimental and autopsy histological studies that show neutrophil infiltration.…”
