Identification of GABRA1 and LAMA2 as new DNA methylation markers in colorectal cancer

Abstract: Abstract. Aberrant methylation of CpG islands in the promoter region of genes is a common epigenetic phenomenon found in early cancers. Therefore conducting genome-scale methylation studies will enhance our understanding of the epigenetic etiology behind carcinogenesis by providing reliable biomarkers for early detection of cancer. To discover novel hypermethylated genes in colorectal cancer by genome-wide search, we first defined a subset of genes epigenetically reactivated in colon cancer cells after treatme… Show more

“…Moreover, the downregulation of LAMA2 expression in human PitNETs could be associated with CpG methylation in the LAMA2 promoter. We have previously reported DNA methylation of the LAMA2 promoter region, 9 which supports the targeting of LAMA2 in various cancers via epigenetic mechanisms.…”

“…Moreover, soluble laminin in culture can suppress cell proliferation in mammary epithelial cells [ 29 ]. DNA methylation at the LAMA2 promoter region found by us and others [ 30 ] suggests that epigenetic mechanisms may target LAMA2 in multiple cancers. These findings suggest the importance of the extracellular matrix during HCC development and perhaps other types of cancers as well.…”

“…Whole-exome and whole-genome sequencing discovered olfactory neuroblastoma and breast cancer has a deletion in LAMA2 21 , 22 . The first exon of LAMA2 is full of CpG islands, which have been reported as hypermethylated in multiple cancers 23 . And promoter hypermethylation is often associated with decreased expression of tumor suppressor genes 24 .…”
