Identification of N‐acetylglucosamine binding residues in Griffonia simplicifolia lectin II

Abstract: Primary structure and crystallographic data of several legume lectins were used to predict the involvement in carbohydrate binding of six amino acid residues (Asp s8, Glu 1°s, Tyr 134, Asn 136, Leu 226 and Gin 227) in Griffonia simpficifofia lectin H (GS-II). The functional involvement of these residues was evaluated by assessing GIcNAc binding of modified forms of GS-II in which these residues were eliminated in truncated peptides or systematically substituted with other amino acids by site-specific mutations… Show more

As GSL-II, like WGA-1 and other GlcNAc-binding lectins, may have a weaker affinity for a single GlcNAc rather than GlcNAc polymers, we may have failed to (secreted)..................................................................... c Relative protein abundance was estimated from the area/height results using SEAQUEST.…”

“…By this technique, several plant lectins, among them of pea (Stubbs et. al., 1986;and Van Eijsden et al, 1992), Erythrina corallodendron (Arango et al, 1993), peanut (Sharma and Surolia, 1994) and Griffonia simplicifolia (Zhu et al, 1996) have been expressed in Escherichia coli. Expression of plant lectins was also achieved in other systems, e.g.…”

“…Both leaf and seed GSII and bacterially expressed rGSII delayed significantly the development of C. maculatus (15). Site-directed mutagenesis was used to identify residues in rGSII essential for GlcNAc-binding activity (33). In the present study, we establish, through bioassays comparing rGSII and variant forms with neutral or GlcNAc-binding null mutations, that GlcNAc binding is required for insecticidal activity of rGSII against the cowpea bruchid.…”
