Identification of microsatellite markers linked to Russian wheat aphid resistance genes Dn4 and Dn6

Abstract: The Russian wheat aphid (RWA), Diuraphis noxia (Mordvilko), is a serious economic pest of wheat worldwide. Host plant resistance is the preferred method to control RWA infestations. The identification and mapping of RWA-resistant genes and the development of resistant wheat cultivars can be facilitated through the use of molecular markers. In the present study, microsatellite (SSR) markers linked to the RWA-resistant genes Dn4 and Dn6 were identified using several F(2) mapping populations derived from crosses … Show more

“…Another RWA resistance gene, Dn4, was mapped on the short arm of the homoeologous wheat chromosome 1D (Ma et al 1998;Liu et al 2002). Dn7 and Dn4 share a common marker, Xksud14 (Ma et al 1998).…”

“…The level of resistance conferred by Dn7 was compared with resistance from two previously characterized genes, Dn4 and Dn2 (Ma et al 1998;Liu et al 2001Liu et al , 2002. '94M370', 'Halt', 'PI262220', and the susceptible cultivars, 'Carson' and 'Gamtoos', were grown in 6-inch pots, with one plant per pot.…”

“…These genes include Dn1, Dn2, Dn4, Dn5, Dn6, Dn7, Dn8, Dn9, and Dnx (Anderson et al, 2003;Botha et al, 1995;Liu et al, 2001Liu et al, , 2002Miller et al, 2001;Myburg et al, 1998). The use of DNA markers in breeding for RWA resistance provides several advantages including the ability to screen year-round instead of only during cooler months of the year when the RWA multiplies, and ability to breed for resistance in places where the aphid does not yet occur.…”

“…Each PCR reaction mixture contained 12.5 μl master mix, 50 ng template DNA, and 150 ng each of left and right flanking primers in a total volume of 25 μl. The PCR conditions were as described by Liu et al (2002). Amplified products were separated in 3% agarose gels at 6 V/cm in 1× TBE buffer.…”
