Identification of Sjögren’s syndrome oral fluid biomarker candidates following high-abundance protein depletion

Abstract: The significance of OF novel depletion methodologies is clearly demonstrated for increased visibility of biomarker candidates as well as for unveiling possible mechanisms involved in this syndrome. This represents a major contribution to our ability to use OF as a future diagnostic fluid.

“…Transgenic mice over-expressing CA I showed aggravated joint inflammation and destruction 6 . CA I has been identified as an oral biomarker in oral fluid of Sjogren's syndrome 7 , and antibodies to CA III and IV have been identified in rheumatoid arthritis 8 . In consideration of the paucity of data on CAs in rheumatic diseases, we have studied the presence of CA IX and XII, two emerging CAs which are pivotal in many human pathologies 4 , in the synovial fluid and synovial cells of JIA patients.…”

“…In another study, the depletion of HAPs such as alpha amylase, albumin, and IgG from oral fluids (OFs) has been reported using alpha amylase affinity device, the ProteoPrep immunoaffinity albumin and IgG depletion kit, respectively . The pooled OF sample (from 18 females patients) and pooled healthy controls (from 18 healthy females) were first subjected to the amylase removing device.…”
