Identification of spinal neurons in the embryonic and larval zebrafish

Abstract: Previous studies indicated that the developing fish spinal cord was a simple system containing a small number of distinguishable neuronal cell types (Eisen et al., Nature 320:269-271, '86; Kuwada, Science, 233:740-746, '86). To verify this we have characterized the cellular anatomy of the spinal cord of developing zebrafish in order to determine the number, identities, and organization of the spinal neurons. Spinal neurons were labeled by intracellular dye injections, application of an axonal tracer dye to all… Show more

“…Previous studies of zebrafish RB neurons have described peripheral processes in the skin (Metcalfe et al, 1990;Bernhardt et al, 1990) but not in the dorsal caudal fin fold. To learn whether this was a characteristic location of RB innervation, we examined fixed embryos labeled with zn-12 antibody (Metcalfe et al, 1990).…”

“…Because anti-islet1/2 antibody is methanol/acetone sensitive, acetone incubation was omitted. RBs in 3d and older embryos were distinguished from other neurons by size, shape, and position in the dorsal spinal cord (Bernhardt et al, 1990). For double labeling with zn-12 or anti-islet1/2 and TUNEL, embryos were first antibody labeled.…”

“…In the zebrafish spinal cord, commissural axons cross the ventral midline and then follow a contralateral diagonal trajectory that is directed anteriorly and dorsally. Upon reaching the dorsal spinal cord, these axons turn to extend along a longitudinal fiber tract (see Bernhardt et al, 1990;Bernhardt, 1994). We have recently used DiI labeling to re-examine the pathfinding of commissural axons on the contralateral side of the midline in the developing mouse and chick spinal cord.…”
